This study used a completely randomized design containing five extract dose treatments (0, 0.5, 1, 2, and 4 g/kg diet doses) and three replications. The undifferentiated Nile tilapia larvae (7 days post hatching) were randomly distributed (n=30) to fifteen aquaria (100×50×50 cm<sup>3</sup>) and maintained for 112 days using a common recirculation system. The results showed that all dose treatments were not significantly different (P&gt;0.05) in gonadosomatic index values of the D84 and D98 samplings. However, the 1 g/kg diet (D112) was significantly different (P&lt;0.05) in all dose treatments. The final histological results (D112) showed that the 1 g/kg diet obtained the highest inhibition level of the testis and ovary developments, which were still in stage II compared to 0.5 g/kg diet (stage III) and control (stage IV and V). The highest average weight, absolute growth rate, and specific growth rate were obtained in the 1 g/kg diet dose which was significantly different (P&lt;0.05) compared to the control. The percentage of males increased significantly (P&lt;0.05) following the increased dose treatment fed to the fish (4 g/kg diet) with 80.12±4.67%, but the survival rate significantly decreased (P&lt;0.05) compared to the control. The administration of 1 g/kg diet dose obtained the best dose and potential as an inhibiting agent for gonad development in Nile tilapia.</p> <p>&nbsp;</p> <p>Keywords: <em>Melastoma malabathricum</em>, gonad inhibition, cytosterol, <em>Oreochromis niloticus</em></p> <p>&nbsp;</p> <p><strong>ABSTRAK</strong></p> <p><strong>&nbsp;</strong></p> <p>Tujuan dilakukan penelitian ini adalah untuk menentukan dosis optimum ekstrak daun melastoma <em>M</em><em>elastoma</em><em> malabathricum</em> yang dapat menghambat perkembangan gonad ikan nila sehingga meningkatkan laju pertumbuhan somatik dan mengevaluasi efektivitasnya sebagai agen seks reversal alami. Penelitian ini menggunakan rancangan acak lengkap yang terdiri atas lima perlakuan (dosis 0; 0,5; 1,0; 2,0; dan 4,0 g/kg pakan) dan tiga ulangan. Larva ikan nila sebelum kelamin terdiferensiasi (7 hari pascatetas) secara acak (n=30) dimasukkan ke dalam 15 buah akuarium (100×50×50 cm<sup>3</sup>) dan dipelihara selama 112 hari pada sistem resirkulasi. Hasil menunjukkan bahwa semua perlakuan tidak berbeda nyata (P&gt;0,05) terhadap nilai GSI pada sampling D84 dan D98. Namun, perlakuan 1 g/kg pakan pada sampling D112 berbeda nyata (P&lt;0,05) dengan semua perlakuan. Hasil histologi terakhir (D112) menunjukkan bahwa perlakuan 1 g/kg pakan mengindikasikan penghambatan perkembangan testis dan ovari yang paling besar yang masing-masing berada pada TKG II, dibandingkan dengan perlakuan 0,5 g/kg pakan (TKG III), dan dibandingkan dengan kontrol (TKG IV dan TKG V). Pengamatan terhadap bobot rata-rata, laju pertumbuhan mutlak, dan laju pertumbuhan harian tertinggi diperoleh pada perlakuan 1 g/kg pakan yang berbeda nyata (P&lt;0,05) dibandingkan dengan kontrol. Persentase jantan meningkat secara signifikan (P&lt;0,05) seiring meningkatnya konsentrasi ekstrak yang mencapai 80,12±4,67% pada perlakuan 4 g/kg pakan, namun tingkat kelangsungan hidup menurun secara signifikan (P&lt;0,05) dibandingkan dengan kontrol. Tujuan dari penelitian ini yaitu untuk mengevaluasi pengaruh suhu dan salinitas terhadap perkembangan, pertumbuhan dan kelangsungan hidup larva siput mata bulan (<em>T</em><em>.</em><em> chrysostomus</em>). Stadia pre-torsion veliger dicapai sekitar 11 jam 36 menit setelah fertilisasi atau sekitar 3 jam setelah trocophor. Stadia post-torsion veliger awal ditandai dengan cangkang yang telah terbentuk sempurna dan pada post-torsion veliger akhir, larva sudah mengembangkan operkulum, kaki, dan propodium. Hasil pengamatan menunjukan bahwa perlakuan A1B3 memberikan waktu pencapaian stadia post-torsion veliger awal dan post-torsion veliger akhir tercepat yaitu masing-masing 19 jam 36 menit dan 22 jam 36 menit setelah pembuahan. Sedangkan perlakuan A1B1 memberikan waktu pencapaian stadia post-torsion veliger awal dan post-torsion veliger akhir terlama yaitu masing-masing 20 jam 30 menit dan 23 jam 25 menit setelah pembuahan. Suhu tidak berpengaruh nyata sedangkan salinitas berpengaruh nyata terhadap laju pertumbuhan harian larva siput mata bulan. Laju pertumbuhan harian tertinggi pada suhu 27±0.5<sup>o</sup>C (A1) tercatat pada perlakuan B3 dan menunjukan nilai yang tidak berbeda nyata dengan perlakuan B2. Suhu dan salinitas memberikan pengaruh yang signifikan namun interaksi keduanya tidak menunjukan pengaruh yang signifikan terhadap tingkat kelangsungan hidup larva siput mata bulan. Perlakuan A1B3 memberikan persentase tingkat kelangsungan hidup tertinggi dan tidak menunjukan nilai yang berbeda nyata dengan perlakuan A1B2. The aim of this study was to evaluate the effect of dietary betaine supplementation on the growth performance and feed utilizationin hybrid grouper (<em>Epinephelus lanceolatus♂ × Epinephelus fuscoguttatus♀</em><em>)</em>. A completely randomized experimental design with four dietary levels of betaine, i.e.0.0%, 0.5%, 1.0%, and 2.0% in quadruplicate was done.Hybrid grouper juvenile obtained from Brackishwater Aquaculture Development Center, Situbondo, with an initial body length and body weight of 5.89 ± 0.05 cm and 2.86 ± 0.09 g, respectively was used as the tested animal. The fish was maintained in 60 cm x 40 cm x 40 cm aquaria with 75 L working capacity with individual recirculating system with a fish density of 15 fish/aquarium for 50 days. Experimental diet was provided to apparent satiation twice a day. The results of this study demonstrated that dietary betaine at a level of 0.5% resulted in higher feed utilization efficiency, protein and methionine retentions, and growth performance and lower ammonia excretion than those of the control (P&lt;0.05). Higher antioxidative status was indicated by the lower malondialdehyde (MDA) in the liver of fish fed with betaine supplemented diets at levels of 1 - 2%. In conclusion, betaine supplementation of 0.5% could increase feed utilization efficiency and growth performance of hybdrid grouper.</p> <p>&nbsp;</p> <p>Keywods: Betaine, Hybrid Grouper, Growth Performance, Feed, Antioxidative Status</p> <p><strong>ABSTRAK</strong></p> <p>Betain memegang beberapa peranan penting dalam pemanfaatan pakan dan metabolisme pada ikan. Penelitian ini bertujuan untuk mengevaluasi pengaruh suplementasi betain pada pakan terhadap kinerja pertumbuhan dan pemanfaatan pakan pada juvenil ikan kerapu hybrid cantang (<em>Epinephelus lanceolatus♂ × Epinephelus fuscoguttatus♀</em><em>). </em>Penelitian menggunakan rancangan acak lengkap dengan empat perlakuan pakan dengan tingkat suplementasi betain yang berbeda, yaitu 0.0%, 0.5%, 1.0%, dan 2.0% dengan empat ulangan. Juvenil ikan kerapu cantang yang berasal dari Balai Pengembangan Budidaya Laut Situbondo dengan panjang dan bobot awal masing-masing 5.89 ± 0.05 cm and 2.86 ± 0.09 g digunakan sebagai hewan uji dalam penelitian ini. Ikan dipelihara dalam akuarium berukuran 60 cm x 40 cm x 40 cm dengan kapasitas 75L yang dilengkapi dengan sistem resirkulasi individu dengan kepadatan 15 ekor per akuarium selama 50 hari. Pakan uji diberikan hingga sekenyangnya dua kali sehari. Hasil penelitian ini menunjukkan bahwa suplementasi betain sebanyak 0.5% menghasilkan pemanfaatan pakan, retensi protein, retensi metionina, kinerja pertumbuhan dan ekskresi ammonia yang lebih baik daripada kontrol (P&lt;0.05). Status antioksidasi yang lebih juga yang ditunjukkan dengan lebih rendahnya konsentrasi malondialdehid (MDA) pada hati ikan yang diberi pakan dengan suplementasi 1-2% betain. NP5 against Streptococcus agalactiae infection https://jma.journal.ipb.ac.id/index.php/jai/article/view/32716 <p><strong>ABSTRACT</strong></p> <p><strong>&nbsp;</strong></p> <p>This study was aimed to evaluate the effectiveness of <em>Bacillus</em> sp. NP5 paraprobiotic administration through commercial feed on growth performance, immune response, and resistance of Nile tilapia against <em>Streptococcus agalactiae </em>infection. <em>Bacillus</em> sp. NP5 paraprobiotic was produced through heat-inactivation at 95°C for 1 h, then performed a viability test on <em>tryptic soy agar</em> (TSA) media and incubated for 24 hours. Paraprobiotics could be used whether the bacteria did not grow on the TSA media. This study used a completely randomized design, containing three treatments with five replications, i.e. 1% (v/w) probiotic addition, 1% (v/w) paraprobiotic addition, and no addition of probiotic or paraprobiotic (control). The experimental fish were reared for 30 days. On day 31 of rearing, fish were challenged with <em>S. agalactiae </em>(10<sup>7</sup> CFU/mL) through intraperitoneal injection route, while the negative control was injected with PBS. This study results significantly improved growth performances and immune responses (P&lt;0.05), compared to control after 30 days of probiotic and paraprobiotic <em>Bacillus</em> sp. NP5 administration. After challenge test, increased immune responses in probiotic and paraprobiotic of <em>Bacillus</em> sp. NP5 treatment had higher survival rates (P&lt;0.05) than positive control. The administration of <em>Bacillus</em> sp. NP5 probiotic and paraprobiotic through commercial feed were effective in increasing growth performance, immune response, and resistance of Nile tilapia against <em>S. agalactiae</em> infection.</p> <p>Keywords: <em>Bacillus</em> sp. NP5 melalui pakan dalam meningkatkan kinerja pertumbuhan, respons imun, dan resistansi ikan nila terhadap infeksi <em>Streptococcus agalactiae</em>. Proses pembuatan bakteri paraprobiotik yaitu <em>Bacillus</em> sp. NP5 diinaktivasi panas pada suhu 95°C selama 1 jam, dilanjutkan dengan pengujian viabilitas dengan menyebarkannya pada media <em>tryptic soy agar </em>kemudian diinkubasi selama 24 jam. Jika bakteri tidak tumbuh, maka paraprobiotik dapat digunakan. Penelitian ini menggunakan rancangan acak lengkap dengan tiga perlakuan dan lima ulangan, yaitu penambahan probiotik 1% (v/w), penambahan paraprobiotik 1% (v/w), dan tanpa penambahan probiotik atau paraprobiotik (kontrol). Ikan perlakuan dipelihara selama 30 hari dan pada hari ke-31, ikan diuji tantang dengan <em>S. agalactiae</em> (10<sup>7</sup> CFU/mL) melalui injeksi intraperitoneal, sementara perlakuan kontrol negatif diinjeksi dengan PBS. Hasil penelitian setelah 30 hari pemberian probiotik dan paraprobiotik <em>Bacillus </em>sp. NP5 menunjukkan kinerja pertumbuhan dan respons imun yang meningkat signifikan (P&lt;0.05) dibandingkan dengan kontrol. Pascauji tantang, peningkatan respons imun pada perlakuan probiotik dan paraprobiotik <em>Bacillus </em>sp. NP5 menunjukkan tingkat kelangsungan hidup yang lebih tinggi (P&lt;0.05) dibandingkan kontrol positif. Pemberian probiotik dan paraprobiotik <em>Bacillus</em> sp. There are four treatments with four replicates per treatment with the use of commercial nano-nutrients (Aquaritin Aquaculture or AA) namely: (1) 0.70 mg/L; (2) 0.525 mg/L, (3) 0.35 mg/L, and (4) without any AA application, but included standard application of using urea and NPK fertilizers to enhance the growth of diatom. The use of AA was successful to trigger the growth of Thalassiosira sp. Group of shrimp treated with 0.70 mg/L had better growth rate. Results of feeding trial indicated that adding AA could also improve the biomass, final mean weight, survival, percentage weight gain, and better feed utilization in terms of FCR. The addition of AA to enhance the growth of Thalassiosira sp. also provides a beneficial impact to the protein composition in whole body of shrimp. Biologically, the protein composition in the whole body of shrimp treated with 0.7 mg/L was higher. The findings from this study showed that the addition of commercial nano-nutrient could enhance the growth of Thalassiosira sp. and led to better growth of shrimp cultured in concrete tank</p> <p>Key words: Growth, Litopenaeus vannamei, nano-nutrients, protein composition, Thalassiosira sp.</p> <p>ABSTRAK</p> <p>Tujuan penelitian yaitu mengevalusi penggunaan campuran unik nano-nutrien untuk meningkatkan pertumbuhan diatom Thalassiosira sp. dan pengaruhnya terhadap kualitas air, laju pertumbuhan, dan komposisi protein pada tubuh udang L. vannamei. Terdapat empat perlakuan dan empat ulangan per perlakuan dengan penggunaan nano-nutrisi komersial (Aquaritin Aquaculture atau AA) yaitu: (1) 0,70 mg/L; (2) 0,525 mg/L; (3) 0,35 mg/L, dan (4) tanpa pemberian AA, tetapi menggunakan penerapan standar pupuk urea dan NPK untuk pertumbuhan diatom. Penggunaan AA berhasil memicu pertumbuhan Thalassiosira sp. Kelompok udang yang diberi perlakuan 0,70 mg/L memiliki laju pertumbuhan yang lebih baik. Hasil uji coba pemberian pakan menunjukkan bahwa penambahan AA juga dapat meningkatkan biomassa, bobot rata-rata akhir, kelangsungan hidup, persentase pertambahan bobot dan pemanfaatan pakan yang lebih baik dalam hal FCR. Penambahan AA untuk meningkatkan pertumbuhan Thalassiosira sp. juga memberikan dampak yang menguntungkan bagi komposisi protein di seluruh tubuh udang. Secara biologis komposisi protein di tubuh udang dengan perlakuan 0,70 mg/L lebih tinggi.
